| Abstract: | Cancer is a complex disease that results from multiple dysfunctions of different mechanism like DNA repair, immune function and apoptotic functions. The term ‘cancer’ is now considered to be a generic name that encompasses more than hundreds of diseases with the same fundamental properties. The disease can be defined by the rapid and indefinite proliferation of abnormal cells in our body. Every year, more than 16 million people are diagnosed with cancer. It was estimated that in 2015, almost 9 million people died from cancer and by 2030, the number might be increased to 11.4million. Cancer is considered as the second leading cause of death worldwide and in most of the cases, death occurs due to an early and inappropriate diagnosis processes. The available common diagnostic methods have comparatively low sensitivity and detect cancer at a delayed stage along with that it brings distress to the patient such as with the use of harmful rays. In this case, biomarkers have been proven to be useful. According to NCI biomarkers are molecule that can be observed in blood or other body fluids and can be estimated as a sign of normal or abnormal process or of a diseased condition like cancer. Long Interspersed Nuclear Element-1 (LINE-1 or L1) is a non LTR retrotransposon, expression of which can be considered as a potential biomarker for a variety of cancers. The reported project is an approach to prepare a specific polyclonal antibody against L1ORF1p- a protein encoded by LINE1. For this LINE1-ORF1 protein was purified and serum was isolated from mice immunized against the purified protein. The specific polyclonal antibody was then tried to be purified from that isolated serum. The specific polyclonal antibody is thought to be helpful in prognosis as well as in the diagnosis of many cancers. Along with this, we were trying to investigate the expression of L1ORF1 in mice testis of different ages; starting from 10-week-old i.e., post puberty. |
